Grapefruit juice increased the plasma concentrations of triazolam, but the sedative effects of triazolam were not enhanced. Pharmacodynamic effects were determined by the digit symbol substitution test (DSST) and utilising a visual analogue scale. The effects of grapefruit juice on triazolam were studied in nine healthy subjects who took 0.25 mg triazolam with or without grapefruit juice. In a randomized crossover study in 10 healthy young subjects, grapefruit juice 250 ml increased the AUC and C max of a single dose of triazolam 0.25 mg about 1.4 times it delayed the t max from 1.6 to 2.5 hours and increased the effects of triazolam slightly, significantly enhancing drowsiness. The half-life of triazolam is prolonged by repeated consumption of grapefruit juice, probably due to inhibition of hepatic CYP3A4 activity. Even one glass of grapefruit juice increased plasma triazolam concentrations, and chronic consumption produced a significantly greater increase. In a randomized, four-phase, crossover study, the effect of grapefruit juice on the metabolism of triazolam interaction was investigated.
